Thank you for your honest assessment of the current longevity movement, and reporting that real scientific breakthroughs remain elusive. What are your thoughts on metformin? As you rightly state, hype often outpaces evidence, and caution is warranted.
Thank you for the helpful reference. I know there are ongoing trials such as TAME (Targeting Aging with Metformin) trial, aimed at testing whether it can be used as a potential anti-aging drug and can delay age-related diseases.
I've sort of peripherally followed the Sirtuin/resveratrol story and resulting acrimony in the field for quite a while, and it has led me to be fairly unimpressed with one of the higher profile people involved in the story... I'm sure you can guess through. I have never seen such a clean timeline of all the relevant details however. Thank you for putting this together. Had not realized you were, to some degree, involved!
As for NAD and related supplements, I have yet to see exciting data that would make me want to recommend this to a patient.
It’s not my area of research, but Matt Kaeberlein thinks that the current evidence does not support that sirtuins help with longevity so it’s interesting that you reference him.
It’s even more interesting that you mention Sinclair given the enormous errors in his research on resveratrol. Fortunately, those errors came out after he sold his company and not before.
NAD+ doesn’t decrease much with aging, though it does decrease because of lack of physical exercise.
Some people think that exercise is beneficial to longevity but they aren’t selling supplements so they are obviously uninformed
It’s a good warning to avoid grifters selling miracle cures… for mice.

I don't think people should take metformin if they don't have diabetes because there is an increased Parkinson's risk in some clinical trial data. You can read more about that here: https://drglorioso.substack.com/p/should-you-take-metformin-if-you

This was really fantastic.
